Could someone please help me. My mask works when I play it using Enter but not when I Test it by pressing CTRL-Enter.

You have a animation inside a group of items and you have added motion to this group. So, can create your motion tween at the main timeline for sync with the rest of the elements at stage. I mean, something like this… (look in the attached file)
